A METHOD OF QUANTIFICATION OF PLANAR CHROMATOGRAPHY RESULTS USING COMMON SOFTWARE

Abstract

Background. The key stage of quantitative planar chromatography is image processing. It’s necessary for further statistic analysis and mixture compound definition. The aim of the study was to offer an express method of thin-layer chromatograms processing and chromatographic curves receiving with available common software.
Material and methods. Flat chromatograms of colored dyes separated on paper sorbent in mixture of organic solvents were taken as the study material. Methodologically the study was based on densitometry of flat chromatograms based on the Lanbert-Beer law about light intensity decreasing, when passing through a thin layer of colored substance. For chromatograms’ image processing the authors used available ImageJ software on the Java platform.
Results. A low-cost express method of planar chromatogram digitizing for quantitative analysis using an office scanner and common software is offered. Important image processing stages are as follows: background subtracting, image grayscaling, brightnes/contrast auto adjustments. A statistical analysis of densitograms’ quality depending on digitizing operations order was carried out.
Conclusions. The quality of densitograms reveals a complex correlation with an image processing operations order. Brightness/contrast auto adjustments either worsen densitogram quality or have a neutral effect. Background subtracting and image grayscaling are important not so much in themselves as in tandem with a background subtracting priority.
